Subject: Proposed Investigation
During my trip this summer I had one idea that seems to be a good one. It is about a proposed method for handling phenylketonuria by injection of nucleic acid.
You will remember that I talked with you last spring about the experiment of Benoit. It had been reported in the newspapers that Benoit, working in the Sorbonne, had injected nucleic acid prepared from one strain of ducks into the abdominal cavity of ducks of another strain; and that, apparently as a consequence of this treatment, the ducks had changed in character, so that they had some of the characters of the strain corresponding to the nucleic acid, although they did not resemble hybrids of the two strains. This phenomenon is somewhat similar to transduction in microorganisms, which has been thoroughly investigated.
While I was in Paris I talked with a number of people about Benoit and his work, although I did not meet Benoit himself. Everyone said that Benoit is a thoroughly reliable man, that he himself was not sure that something might not have gone wrong in his work, that he regretted not having carried out any controls, but that his reported results should be accepted at their face value. I visited the Macromolecule Laboratory of Sadron in Strasbourg, and talked with the young woman who had prepared the nucleic acid. Then, when I returned to Paris, I found that the newspapers were carrying an account of a later report by Benoit, made to the Academy of Sciences in Paris. Benoit reported that ducklings from the changed ducks and drake had inherited the changed characters.
The idea that occurred to me at about this time is that it might be possible to introduce into the cells of a phenylketonuria child some nucleic acid that would be able to carry out the synthesis of the enzyme that is lacking in the patient, and that in this way the disease might be brought under control.
I think that the proposed treatment probably should be carried out with use of nucleic acid made from sperm of the father of the child; at any rate if this is possible. One reason for this is that this nucleic acid would introduce fewer characters that are foreign to the child itself than nucleic acid from another source. If all genes were heterozygous in the father, the child would have inherited just half of the father's genes, and the other half would be foreign. Only one of the two phenylalanine oxidase genes would be useful, however, so that the ratio of useful phenylalanine oxidase gene to foreign gene would be just the same as for nucleic acid from an unrelated source. However, probably many, perhaps most, of the genes in the father are homozygous, so that neither one of a pair would be foreign to the child. If, for example, 90 percent of the genes were homozygous, the number of foreign characters introduced per effective gene with use of nucleic acid from the father would be only 20 percent as great as with use of DNA from another source. There is, also, probably an emotional or psychological value to using DNA from the father.
If the decision is made that it would not be improper to carry out this proposed therapy before animal experiments have been tried, I suggest that we plan to go ahead at once. Presumably the greatest probability of success would come in the treatment of very young patients, but it might not be necessary to restrict the trials to these patients.
I suggest also that we plan to begin some animal work immediately. I do not know what inborn error of heredity in animals would be best suited to these studies, but there are without doubt some such errors known that would be so suited, and I request that you make a literature search, in order that the work could be started at once. If an experimental error with a recessive inborn heredity could be found, there would be the possibility of carrying out extensive studies of this therapeutic method.
The proposed therapy of injection of DNA could, of course, be tried also for other diseases involving abnormal genes. If a galactosemic child is available, it might be worth while to apply the proposed treatment in this case too.

Dear Mamma,
I have not written to you for ages but have been thinking about you on your travels, and
should love to hear about your experiences in Russia whether the celebrated Pauling charm
worked irrespective on the iron curtain. Peter once remarked on looking at a picture of a bunch
of assorted diplomats in a newspaper that the Russian looked, at least, far the friendliest.
We heard via Barbara Dunitz's grapevine that Linda had been visiting Honolulu a while
ago. I hope this visit was not for health reasons- you told me she had not been feeling too well
before you left. I don't know her new address properly.
We have been having rather a straining time recently. A common friend of Peter and
Linda, who is subject to nervous breakdowns; came and recuperated with us after the latest of
these! Hers is a very deep and I think insolvable problem, and it was extremely depressing and
exhausting to try to help her through this. I think we recuperated her, physically at least, but she
left me a nervous wreck. She arrived in what she described as a hypermanic state. Which
caused her to talk solidly for ten days, only stopping to eat, which she needed to do frequently
and copiously to supply her with energy to keep on talking. So I was almost run to a shadow just
by supplying victuals.
Thomas was also a little upset by all this but settled down and was nice to her at the end.
He is teething again and grinds his six teeth constantly which is horrible to hear. He had a
birthday party for four little friends but it was a sad failure, he sat in a corner and cried and was
only comforted by much cake and many toys: We gave him a red train and some stacking tins.
Talking of presents, I don't think I ever thanked you for my birthday present, but please forgive
me, and I send now accumulated thanks with interest. It is safely tucked away in Peters account
(if that is called safe).

I had intended to have a chat with you, when I saw you in Cambridge this summer, about the three hydrogen bonds between guanine and cytosine, but I forgot to do so. Now I have been jogged into thinking about the matter again by reading your article in The Scientific American.
I suppose that you saw the paper that Professor Corey and I published in Archives of Biochemistry and Biophysics last year, in which the matter is discussed. I enclose a reprint.
It seems to me that there is no justification whatever for continuing to show guanine and cytosine with only two hydrogen bonds, as you do in The Scientific American article, both in the text and in the figure. What do you have to say?

In answer to your letter of 3 September, I send you herewith a copy of my paper "The Molecular Basis of Genetics," which covers the subject matter of the address that I gave at the 112th Annual Meeting of the American Psychiatric Association in Chicago last year.
You will note that I start out with the statement "I am sure that most mental disease is chemical in origin..."
I gave my address without use of a manuscript, and I am sure that it was not identical with the published paper. I do not remember exactly what I said, but I may have made some reference to the work of Dr. Heath of Tulane University Medical School. Dr. Heath has published an account of his experiments on the injection into normal human beings of blood from schizophrenic individuals. He reported some peculiarities in behavior of the recipients. The reported effects, which have been attributed to a substance taraxein presumed to be present in the blood of the donors, were only transitory. I am not sure how much significance is to be given to these experiments, and I am confident that I did not make the statement that if blood from a schizophrenic were transfused into a normal person, the normal person would have schizophrenia in all its manifestations.
In answer to Mr. Greer's question, I would say that I doubt that any schizophrenia is being dispersed through the blood banks. This is not a problem at all comparable to that of, say, dispersing hepatitis virus.

I'm very sorry about this Scientific American article. It shouldn't have happened, though it wasn't entirely my fault.
As to the three bonds, I had your reprint on my desk when you were here in July. I didn't mention it to you because I was completely in agreement with your conclusions. It seems a good general rule that a C = O or an NH will almost always make a hydrogen bond, either to another molecule or to water. If it is not making a bond and if there is no room for a water molecule something is likely to be wrong. We have been using this idea in our collagen models.
I was not responsible for producing the figures for the Scientific American article, nor for the legends to the figures. They were sent here for my approval only two days before they went to press. I was very annoyed at this, especially with the clumsy stereochemistry. I did cable a few alterations to the legends, but it seemed too difficult to get the figures altered satisfactorily by cable. In my haste I quite overlooked the offending legend.
The next time I have to write about DNA I will include a reference to your paper, and mention the error in the Scientific American article.
Things are quite hectic here. Americans seem to join the unit every day. John has a very nice three-dimensional Fourier of myoglobin (out to 6 A) which is very encouraging.

Dear Fred:
I have found your two letters waiting for me after returning home from our summer in Europe. Ava Helen and I were gone nearly three months, half of it in France, and the rest in England, Germany, Norway, Yugoslavia (one week), and Russia (three weeks). We had a great time - I must say that it was interesting to see what life is like behind the iron curtain. I am glad to be home, however.
Linda was married a couple of weeks ago. She married Barclay Ray, Professor of Geology here, and she and Barclay are now finishing up their honeymoon, which they spent at our ranch. Ava Helen and I have just been to the ranch. I don't know whether you know that we have a ranch. We bought it about a year ago. It is a quarter section, which was homesteaded by a local man in 1916, so that we are the second owners. It is on the coast, 19 miles north of San Simeon, and extends from sea level (about a half a mile of beach) to about 2000 feet, or perhaps only 1500 feet - the mountains keep rising after the boundary line is reached. We have a little cabin there, 2 rooms, and a barn, and there are 15 cows grazing there at present.
I am glad to hear about the two raises that have come your way. With respect to the old memory not being too reliable, I am afraid that I have to admit the same thing. I am having an especially hard time in remembering the names of people.
Crellin and his wife and daughter left for Portland a few days ago. I don't know whether you knew that he was married last summer, to a Pasadena girl who was attending Reed. Their baby was born this summer. We saw Peter and his wife and little Thomas early in July, when we visited London. He expects to have his Ph.D. work finished up by the end of this academic year - he is working with Professor Nyholm in the field of inorganic chemistry, University of London. Linus came over from Honolulu for Linda's wedding. We were glad to see him again. He and his wife have built a house in Honolulu, so I judge that they intend to stay there for sometime.
Professor Corey is apparently pretty well pleased with the way that the protein project is going. Linda has been working on it for several months, and she succeeded in growing some crystals of lysozyme with the columbium complex included, and also with the corresponding tantalum complex. I think that she will continue work after she and her husband settle down to married life, and I hope that she continues to be successful with the crystal growing job.
Our research project on mental disease is making rather slow progress, but I suppose that we have to consider the progress satisfactory. We have not worked very much on anything except mental deficiency, and I do not have any special information about the various new drugs that are used for mental illness. If I hear about anything especially hopeful I shall let you know.

Dear Peter:
I am pleased to hear about the 6 Å three-dimensional Fourier of myoglobin that Kendrew has made. The same description of it had already reached Pasadena, through Dintzis. We are eager to learn the details.
I hope that you are getting along well with your cobalt sulfate structures. This is, of course, the most important activity for you, but I should like to hear also about any progress that you make on the radial distribution curve for water. I am pretty busy, trying to get caught up in the work that has piled up for me, but, on the other hand, I should like to get the paper on water published.
The wedding went along fine. Linda was beautiful and Barclay was handsome, the mother and the father of the bride were about as expected, much champagne was drunk by the guests, and in general everything went along well. I think that Linda and Barclay had a fine time at the ranch on their honeymoon. Mamma and I have just been up there for two weeks. There are various problems around the laboratory that I have to take care of.
Linus turned up at about noon on the wedding day and left again at 11 o'clock that night. Perhaps he has come to see you - I think he was going to spend a few days in Europe. He is looking well. Crellin and Lucy and the baby seem to be well also, although Crellin was down with a cold, and he and Lucy had to postpone their departure for Portland by a couple of days.
I think that Linda will go back to work for Professor Corey. He seems to be well satisfied with her as a crystal grower, and is hoping to have her back on the job. Barclay and Linda have rented the house on Barhite Street from Mrs. Jordan, and they have moved in there.
