I thank you very much for your kindness in sending me a copy of your book La Mineria y la Metalurgie en la America Espanola durante la epoca colonial, which I have examined with much interest.
In particular, I was pleased to see the detailed discussion that you give of the outstanding work of Don Andres Manuel del Rio y Fernandez and his discovery of erythronium in 1801.
It seems to me that the effort should be made to get the name of erythronium adopted officially by the International Union of Pure and Applied Chemistry. I have not had an opportunity to examine the original papers of del Rio, but I judge that there is no doubt that he has priority, by nearly 30 years.
If you decide to raise the question again with the International Union of Pure and Applied Chemistry, I should be glad to join with you in the effort.

I am writing to tell you about my situation with respect to research, and about the disposal of the $50,000 fund.
At the present time there remains unallocated the sum of $7735.10. A large part of the allocated sum has not yet been expended-only $2062.50 has been expended so far in salaries (for Dr. Murayama, who is assisting me on work on hemoglobin). The remaining salary items are for men whom I have appointed for the coming year, including Dr. Murayama. The other three have not yet taken up residence here.

I am especially in two research programs at the present time, both of them in their beginning stages. I think that it will be possible within a few years to make a complete structure determination of a crystalline globular protein, such as hemoglobin, insulin, ribonuclease, etc. This problem is an extremely difficult one. The hemoglobin molecule, with molecular weight 68,000, contains about 7,500 atoms other than hydrogen, and the job of making a complete structure determination involves determining three cartesian coordinates for each of these atoms. X-ray techniques have not in the past been powerful enough to permit such a job to be done. I believe that we have discovered a way of doing the job, provided that it is worked at intensively, for a long time-several years. The first step in making use of our new method is to grow crystals of a protein and two inorganic complexes, one a cation, such as Ta6Co12
++ and the other an anion, such as W12PO40
---. There is only one man in the world who has had much experience in attacking the problem of growing crystals of this sort. He is Dr. Jacques Lewin, who is assistant director of the Centra National de Transfusion Sanguine, in Paris. I have written to him, offering him appointment for one year, as Senior Fellow in Research. I hope that he will be interested to come to Pasadena, and that he can obtain leave from his post in Paris for this purpose. We should be able to learn something about his special techniques during his visit here, and he himself might, in the course of one year, be able to grow just the crystals that we need to begin our attack on this problem.
Professor Herbert Jehle is Professor of Theoretical Physics at the University of Nebraska. He has been working for some years on the quantum mechanical theory of specific forces operating between proteins and other biological molecules. I have been interested in this field too, and have invited him to come here for one year, in order that we might collaborate. The University of Nebraska is giving him part of his salary, $2500, for the year, permitting him to accept our offer. I am hoping also to get him to work on some theoretical aspects of our method of determining the structure of globular proteins.
Dr. Murayama is working on the structure of proteins in relation to mental disease. He is at present studying hemoglobin obtained from the blood of patients with phenyl pyruvic oligophrenia. I expect that Dr. Helfferich, when he arrives in Pasadena, will also collaborate in the work on mental deficiency.
I may say that it may be possible to transfer some of these men to other funds. I have applied, as you know, to the Ford Foundation for a grant for support of our work on mental deficiency. The Ford Foundation has not yet reported its decision on my application. The Langley Porter Clinic, in San Francisco made four applications, referring to four different projects that they proposed, and they have been notified already that all four of their applications have been turned down. I think that it may be that the Ford Foundation is planning to concentrate its support on private institutions. At any rate, I am hoping that they will act favorably on my application.
Also, we have applied to the Navy for a contract to permit an intensive attack on the problem of the determination of the structure of crystalline globular proteins. This is really a great problem-it has the possibility of leading to great advance in our understanding of the human body, and to significant contributions to biology and medicine. It is a problem that can be solved only by an intensive attack over a long period of years. Now that I have had my 55th birthday, I am thinking about how long I have remaining to me, for effective scientific work, and I suppose that I should be thinking about 10 years as a period of time in which I must do what I want to do. I think that the problem of the structure of globular proteins can be solved within 10 years; perhaps one complete structure determination can be made within 5 years, if we are able to attack the problem vigorously enough. It would not be enough, of course, to determine the structure of a single protein-if the structure of hemoglobin could be determined, one would still want to know the structure of ribonuclease, pepsin, trypsin, or some other enzyme (perhaps of several enzymes), and of insulin or of some other hormone (perhaps of several)-I suppose that the physiologists, biochemists, and medical men would not be satisfied until complete structure determinations had been made of a rather large number of protein molecules. The mental deficiency project is different in nature. There is almost infinite possibility for making progress in the attack on this problem.
Mama and I are getting along well. We are making plans for entertaining Professor Hugo Theorell, and his wife, week after next. Professor Theorell was the recipient of the 1955 Nobel Prize for Medicine. He is an old friend of ours-he came to work in our laboratory in 1939 or 1940. You may remember him from that time-a rather stocky man badly crippled by polio, so that he walks with difficulty with the aid of two canes.
Mama and I are making an eastern trip. We shall stop in Portland for one day, in Vancouver, and then go to Winnipeg, where I am to give a lecture in the Medical School. Then we go to the University of Illinois, Yale, Amherst, and home. I am giving an address at the annual meeting of the American Psychiatric Association in Chicago.
Also, we are planning to make a short trip to Europe during June. I have been invited to deliver the Avogadro Commemoration Address in Rome.

SUBJECT Proposed calculation, relating to crystalline globular proteins
I suggest that you have Dr. Marsh and assistants carry out a calculation as a test of methods of determining the structure of crystalline globular proteins.
The simple calculation involves the formulation of a one-dimensional structure, the calculation of the real and the imaginary parts of the amplitude of the scattered wave, A and B, and the use of these values, extending over various ranges of interatomic distances, to evaluate various Fourier series, in order to see what ranges of interatomic distances must be used to get resolution of atoms and identification of atoms.
I suggest that the work be done, as a first step, roughly in the following way. Let us take a one-dimensional structure, with a0 = 30 A. The unit is to have one Fe at 0.000, one Cl at 0.210, and 12 C at 0.080, 0.125, 0.290, 0.370, 0.420, 0.470, 0.540, 0.630, 0.700, 0.750, 0.840, and 0.900. These coordinates make interatomic distances along the line cover the range from 1.35 A to 3.00 A. I suggest that A and B be calculated for orders h from 1 to 30, representing interplanar distances from 30 A to 1 A. The f value of carbon might be used as the basis of the calculation; I think that it would be good enough to take that for chlorine as 3 times that for carbon, and for iron as 5 times that for carbon.
These values of A and B could then be used to evaluate the Fourier series representing the electron density, by a number of different calculations, corresponding to use of orders of reflection as follows: 1 to 30; 6 to 30; 8 to 30; 10 to 30; 15 to 30; 10 to 20; 10 to 15; others indicated by the results.
The Fourier series obtained in this way could be discussed with respect to errors in the coordinates of each atom, and also in the height of the peak representing each atom. What limitation of the range causes an atom to be missed? What limitation of the range leads to the introduction of phantom atoms - peaks in positions where no atom was present?
After these questions have been answered, a suitable range may be selected, perhaps 10 to 30, reflections from 3 A to 1 A spacing, and the calculation repeated after random 10% errors have been introduced into the values of A and B.
Probably the next step that will have to be made is to carry out a similar calculation for a two-dimensional structure, involving about 100 atoms. It might be possible to generalize from the results of the one-dimensional problem and the two-dimensional problem to conclusions valid for the three-dimensional problem; but it probably will be worth while to carry out some sort of detailed theoretical treatment of this sort also for the three-dimensional problem.

I note that I have forgotten one item, in reporting to you about the expenditures on the $50,000 fund.
I have allocated $1200 of the fund as the salary for 3 months for Professor A. Christomanos.
Professor Christomanos is the Professor of Chemistry in the University of Athens. He came here in September 1955, as the recipient of a Smith-Mundt Fellowship, with the plan of carrying on research in the field of Chromatographic analysis of proteins. I recommend that, after a preliminary period of devoted to learning the techniques of amino-acid chromatography from Dr. Walter A. Schroeder of our laboratories, he attack the problem of the nature of the end groups of the hemoglobin molecules in the blood of patients with phenylpyruvic oligophrenia. He has now begun work on this problem, which may throw some light on the nature of this kind of mental deficiency. The idea is that the presence of a large amount of phenylalanine in the body fluids of patients with this disease may cause abnormalities in the amino-acid composition of protein molecules, and Professor Christomanos will examine the amino acids at the free amino end of the polypeptide chains, and perhaps the adjacent ones also, to see whether a phenylalanine residue gets into these positions by mistake. Normally valine is present at the end of the polypeptide chains, and leucine in the second position.
Dr. Christomanos has been effective in his work and wants to stay longer than the period for which his Smith-Mundt was originally granted. He applied for an extension, and was given a two months' extension, for the months of April and May. I then told him that we would give him an appointment for three more months, ending August 31, 1956. He should be able to do a good job on his problem in this time.
Of course, if the Ford Foundation grant is made, I shall be able to transfer him to this budget.
Mama and I are planning to spend the month of June in Europe. We probably shall leave Pasadena, on SAS, at midnight on Wednesday 30 May, flying from Copenhagen to London on Friday 1 June, and staying until Monday 4 June with Peter and Julia. Then on Monday we shall fly to Rome, in order that I may give the Avogadro Commemoration Lecture for the Academia dei Lincei. We shall stay in Italy until about 20 June. I don't know what we shall do then-perhaps return to London for a week, go to Copenhagen for a few days, and fly home early in July.
Are you (Anita) going to stop to visit us on your way to Europe? We shall be away from Pasadena for some time, but shall be at home from 14 to 18 April, and from 9 to 30 May. Then we shall probably be home again after 8 July. I shall be busy during the summer in connection with my research programs, and I also hope to work on a new edition of THE NATURE OF THE CHEMICAL BOND.

Professor Corey and I, while thinking about our work during the coming year, have decided that we need to have advice. Will you write a few pages to me, answering the following questions?
You know that only a little work has been done on crystalline globular proteins in our laboratory, and that we may well have used rather amateurish methods. During the coming year we hope to carry out a preliminary investigation, involving determination of the size of the unit and of the space group, for a considerable number of protein crystals, and we should like to do this preliminary work in the most expeditious manner. Would you be good enough to tell me how this job is done in Cambridge, and also to add any other information that you think pertinent? How do you grow the crystals? How do you mount them? What procedure do you follow in getting preliminary x-ray information, about size of unit and space group? Do you make use, for this purpose, primarily of precession cameras? If so, what dimensions are they, and what manufacturer? Can you get all of the information that you need from a single crystalline specimen, without changing its mounting? How do you determine the orientation of the crystal? How long does it take you to obtain this preliminary x-ray information, after you have got a good single crystal? So you make use of rotation photographs, or of Weissenberg photographs, also? If so, what sort of cameras do you use - what radius, etc.?
Mama and I shall leave on 23 March, flying to Portland, then to Vancouver by night, from Vancouver by Canadian National to Winnipeg, where we arrive on Tuesday 27 March. We shall be in Winnipeg, care of the Department of Chemistry, until Saturday 31 March. We then stop for two days in Iowa, and on 4 April arrive in the University of Illinois, address Chemistry Department. We shall be in Illinois, with a short trip home from 14 to 18 April and a visit to Yale and Amherst the week of 23 April, until 5 May, and then return home, stopping for a day in Lawrence, Kansas. We then leave Los Angeles for Copenhagen on the night of Wednesday, 30 May, arriving the afternoon of Friday 1 June in London, by SK 501, at 1:15 P.M. We leave for Rome on Monday 4 June, about noon. We expect then to return to London about 20 June, for another week, to go to Copenhagen for a few days, and on home. The W.H. Freeman Company is celebrating its tenth anniversary on 6 and 7 July, and we are rather committed to be there.

Dear Dr. Kuffner:
I have been interested to read your letter of 9 March and the accompanying material, dealing with philosophy in relation to science.
I may say that I myself have had trouble in getting interested in philosophy. From time to time I have made an effort to do so, but never with success. During recent years I have talked with Dr. Max Mason about this matter, and have found that his thoughts on the subject are similar to mine. Perhaps he goes somewhat beyond me - he says that so far as he can discover philosophy is verbalism - that it deals with words exclusively, and has no deeper meaning. It is my opinion that there is something to what he says; and I am skeptical about the value of philosophy for young people. Of course, some of the questions that you mention are really a part of science, a clarification of the words used by scientists and of the questions that they discuss.

There is described in Nature of 3 March 1956, near the middle, a set of four magnifiers, 6X, 8X, 10X, and 15X, in a silk-lined case, to be sold for 77 shillings 9 pence. The company is Newbold and Bulford, Ltd., Enbeeco House, Rogers Street, Grays Inn Road, W. C. 1.
The Theorells hare been visiting us, for four days. They leave tonight for home, by SAS. Yesterday Mama and I took them, in the Riley, to Palomar, and home by way of Oceanside and Laguna. We had a dinner for them on Monday, with Max Mason and Mrs. Mason, Ruth Tolman, and some other people as guests. Tonight we shall have a reception before taking them to the plane.
